Test performed on whole human skin explant. The effects of the product on the epidermis are evaluated by an MTT cell viability test ( 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) cell viability test).
The Prediskin assay has been designed by Eurosafe in the early 2000s for irritancy testing of finished cosmetics: for ingredients, the assays to perform are those of the OECD test guidelines whereas there is no official guidelines for the finished products.
The assay relies on discs of native human skin that are cosmetic and plastic surgery leftovers, collected in full regulatory compliance in matter of human products donated for research purposes.
The test product is applied on the discs during 20h and viability is measured with a MTT assay.
The classification rule gives a not irritant/ irritant score, and the test formulation is benchmarked to products in the same cosmetic category/ies.
The Prediskin assay can be considered as a in vitro patch testing assay.
